Kisspeptin Peptide: What It Is, Dosage and Testosterone

What is kisspeptin, and does it raise testosterone? Kisspeptin is the brain signal at the top of your testosterone chain: it starts the hormone relay (GnRH, then LH and FSH) that tells the testes to make testosterone. In small human studies it raised LH and testosterone in men, but frequent dosing can dull the response. Most Regimen users on kisspeptin take about 200 mcg a shot, daily or a few times a week.

Kisspeptin sits at the very top of the testosterone chain. It is the signal that tells the brain to release GnRH, which tells the pituitary to release LH and FSH, which tell the testes to make testosterone. It works on your own system rather than replacing testosterone, which is the main difference from TRT.

  • What it is: Peptides from the KISS1 gene; kisspeptin-10 (10 amino acids) is the one sold
  • FDA status: Never approved; kisspeptin-10 is on the FDA’s compounding safety-risk list
  • Sport: Banned at all times in men by WADA (S2.2.1)
  • Human studies: Small trials: LH and testosterone rise, IVF egg maturation
  • What people take: 200-330 mcg a shot (Regimen users), 200 mcg most common
  • Common vial: 10 mg, mixed with 2 or 3 mL
  • Half-life: About 4 minutes (kisspeptin-10), 28 minutes (kisspeptin-54)

What Is Kisspeptin?

Kisspeptins are a family of peptides made from one gene, KISS1. The body cuts the same starting protein into kisspeptin-54, -14, -13 and -10, which share the same last 10 amino acids. Kisspeptin-54 is the main form in human blood. Kisspeptin-10 is the shortest piece that still works fully, and it is the form sold as a peptide.

It acts on a receptor in the brain (GPR54, also called KISS1R) and releases the brain’s own store of GnRH. GnRH then drives LH and FSH from the pituitary, and LH drives testosterone. Kisspeptin is needed for puberty, and it was first found as a gene that held back the spread of cancer cells, which is why the 54-amino-acid form is also called metastin.

Does Kisspeptin Raise Testosterone?

In men, short-term studies say yes:

  • Kisspeptin-54 by IV (2005): in 6 healthy men, LH more than doubled and testosterone rose about 15%.
  • Kisspeptin-10 by IV (2011): a single dose raised LH, and a 22.5-hour infusion raised testosterone about 45% in 4 men.
  • Men with type 2 diabetes and low testosterone (2013): an infusion raised testosterone by about a third.
  • Infusions under the skin (2026): in 7 men, 8 hours on and 16 hours off kept LH, FSH and testosterone up for 12 days.

How often you dose matters. In women with amenorrhea (no periods) given kisspeptin-54 twice a day, the LH response had almost gone by day 14 (from 24.0 to 2.5 IU/L). Twice-weekly dosing only partly dulled it. And in men on a continuous 5-day infusion, LH and FSH fell back to placebo levels, though testosterone stayed raised. More often is not better.

Kisspeptin Dosage (Kisspeptin-10): How Much and How Often

There is no approved kisspeptin-10 dosage. The human studies dosed by body weight, mostly by IV:

  • Kisspeptin-10 by IV: the LH response peaked at 1 mcg per kg, and 3 mcg per kg gave a smaller response.

What people take: most Regimen users on kisspeptin take 200 to 330 mcg a shot, and 200 mcg is the most common dose. 39% take it every day and 30% three days a week; most of the rest take it every other day or twice a week.

How to Reconstitute Kisspeptin (Units to Draw)

73% of Regimen users on kisspeptin use a 10 mg vial, and most mix it with 3 or 2 mL of bacteriostatic water. Kisspeptin dosage chart, in units on a 100-unit (1 mL) insulin syringe:

Dose10 mg + 3 mL10 mg + 2 mL
100 mcg3 units2 units
200 mcg6 units4 units
300 mcg9 units6 units

With a 5 mg vial and 2 mL, 200 mcg is 8 units. At 2 to 6 units, a small misread is a big share of the dose. More water means more units for the same dose, which makes small doses easier to measure. For any other mix, use the peptide calculator.

Kisspeptin Side Effects

In the human studies, kisspeptin was well tolerated: the recent trials in men and women with low sexual desire reported no side effects. In an IVF trial in women at high risk of ovarian overstimulation, no one developed moderate or severe overstimulation.

These were small, short studies. The FDA put kisspeptin-10 on its compounding safety-risk list (Category 2) in 2023, saying it may cause immune reactions by some routes, can be hard to make without impurities, and has little safety data for the proposed routes. Long-term effects are not known.

Kisspeptin vs TRT, hCG and PT-141

  • TRT supplies testosterone from outside. See the testosterone cypionate dosage guide.
  • hCG acts like LH directly on the testes. See hCG for men on TRT.
  • PT-141 (bremelanotide) works on melanocortin receptors in the brain, not on hormones. As Vyleesi, it is FDA-approved for low sexual desire in premenopausal women.
  • Kisspeptin works further up, in the brain, so the whole chain from GnRH down runs on your own hormones. Unlike TRT and hCG, it isn’t an approved drug.

Among Regimen users on kisspeptin, 24% also track testosterone, 23% PT-141 and 16% hCG.

Kisspeptin Benefits for Fertility and Libido

In IVF trials, a single shot of kisspeptin-54 under the skin (1.6 to 12.8 nmol per kg) triggered egg maturation: in 60 women at high risk of overstimulation, 95% had mature eggs. A second dose 10 hours later raised the share of women with a good egg yield from 45% to 71%. In women early in their cycle, kisspeptin-10 did not raise LH even at high doses. Kisspeptin also changed how the brain processes sexual cues in small studies of men and women with low sexual desire.

How Long to Run Kisspeptin

34% of Regimen users on kisspeptin run it in cycles, most often 4 or 8 weeks on with 2 to 4 weeks off.

Where to Inject Kisspeptin

Kisspeptin goes under the skin. Most Regimen users on kisspeptin inject in the belly or the thigh. The peptide injection sites guide shows each spot.

Kisspeptin has never been approved by the FDA, and kisspeptin-10 is still on the FDA’s compounding safety-risk list (Category 2), so US compounding pharmacies are not supposed to make it. The 2026 WADA Prohibited List names kisspeptin and its analogues among testosterone-stimulating peptides banned at all times in men (S2.2.1).

Track Kisspeptin With Regimen

Kisspeptin is about what happens downstream, so the numbers that matter are on your bloodwork. Regimen, a peptide tracker, puts testosterone, estradiol (E2) and SHBG, scanned or typed in, on the same timeline as your doses; the TRT blood work guide covers what to test.

Frequently asked questions

Does kisspeptin increase testosterone?

In men, yes, in the short term. In small studies, kisspeptin raised LH and testosterone within hours, including in men with type 2 diabetes and low testosterone. Given continuously or very often, the LH response fades.

How much kisspeptin should a man take?

There is no approved dose. Human studies used weight-based doses, mostly by IV. Most Regimen users on kisspeptin take 200 to 330 mcg a shot under the skin, and 200 mcg is the most common dose.

How often should I inject kisspeptin?

No schedule has been tested for long-term use. In studies, twice-daily shots dulled the response within two weeks, while twice-weekly shots only partly did. Among Regimen users, 39% take it every day and 30% three days a week.

How long can you stay on kisspeptin?

The human studies lasted from a single dose to a few weeks. 34% of Regimen users on kisspeptin run it in cycles, most often 4 or 8 weeks on with 2 to 4 weeks off.

Is kisspeptin better than TRT?

They do different jobs. TRT supplies testosterone from outside. Kisspeptin asks your brain to make your body produce its own, and it is unapproved, with only short-term studies behind it.

Can I take kisspeptin and testosterone together?

People do: 24% of Regimen users on kisspeptin also track testosterone, and 16% track hCG. No study has tested kisspeptin together with TRT.

Is kisspeptin worth taking?

It depends on the goal. Short studies show it raises LH and testosterone in men and helps eggs mature in IVF, but it isn't approved and there are no long-term studies.
